Maintenance of discrete bacterial populations in adjacent marine habitats. ArchiMer

Some mechanisms of selection which operate on marine Vibrio and Pseudomonas strains during survival in dilute environments have been examined. Mannitol uptake and utilisation systems are more efficient in the Vibrio strain compared with the Pseudomonas strain during starvation survival. A Vibrio mutant strain lacking mannitol uptake showed no greater loss of viability during starvation survival than mannitol positive strains. The Vibrio strain appears to be constitutive with respect to mannitol uptake systems, whereas the Pseudomonas strain examined is inducible. Isolement et identification de vibrions halophiles d'une station experimentale de mariculture ArchiMer

110 Vibrio strains were isolated, at various times of the year, from samples of sea-water supply, breeding-tank water and discarded fish used as food at the breeding stations. From the results obtained by the biochemical and cultural tests, it was possible to identify most of the isolated strains as Vibrio anguillarum . Strains with intermediate characteristics, above-all positive saccharose, proved to be numerous. The authors discuss the occurrence, in particular environments such as those being studied, of atypical or intermediate vibrio strains which are difficult to classify.

Bacteria associated with semi-continuous culture of Brachionus plicatilis , which is used as the first food of marine fish larvae, were counted on Zobell 2216 E medium and identified. The numbers of bacteria were very high in both Brachionus (about 10 super(11) bact. per ml of digestive content) and in water (from 1.6.10 super(6) to 4.7.10 super(7) bact. per ml). The cultures were studied at driving a time when mortalities occurred frequently. Water and sediment samples monthly collected from Arcachon Bay (Gironde, France) were examined during three years for the presence of marine vibrios. V. cholerae serotype 01 was never detected. On the other hand, the Eyre River and mouth contain non-agglutinating V. cholerae (127 strains) principally in water. The low temperatures slow their development down without stopping it completely. V. parahaemolyticus (257 strains) present in the same areas is particularly isolated from water when the aquatic medium temperature rises to 15 degree C. Bacterial problems associated with scallop Pecten maximus larval culture ArchiMer

Scallop Pecten maximus larvae have been cultured at the Argenton and Tinduff (Brittany, France) hatcheries with antibiotic treatment (chloramphenicol at 8 ppm) for 15 yr. Without treatment, outbreak of disease has normally occurred between Day 12 and Day 19 or sometimes earlier. A bacteriological study of larvae reared with and without antibiotic was performed over a 4 yr period. Among the collected strains, 2 clusters (C and F) of vibrios were present at high densities only in larvae cultured without treatment. Vibriosis is a major disease problem in shrimp aquaculture. 'Syndrome 93' is a seasonal juvenile vibriosis caused by Vibrio penaeicida which affects Litopenaeus stylirostris in grow-out ponds in New Caledonia. This study assessed the toxic activities of extracellular products (ECPs) from V: penaeicida, V. alginolyticus and V, nigripulchritudo using in vivo injections in healthy juvenile L. stylirostris (= Penaeus stylirostris) and in vitro assays on shrimp primary cell cultures and the fish cell line epithelioma papulosum cyprini (EPC). Recherche et identification de bactéries pathogènes au sein d'une écloserie nurserie d'huître creuse, Crassostrea gigas, en relation avec la mortalité estivale du naissain ArchiMer

Summer mortality has been observed on oyster spat Crassostrea gigas since 1991 on french coasts. In our study, we deseribed symptoms observed during the mortality, especially adductor muscle weakness and we isolated strains on Zobell medium and TCBS from moribund spat. Strains isolated during a high mortality (60 %) were selected by experimental pathology on spat. Inoculation by bath led us to select a pathogenic strain (TNEMF6) for oyster spat. Phenotypic identification, phylogenetic study on 16S rDNA and quantitative DNA/DNA hybridisation showed that this strain belongs to Vibrio splendidus biovar II. Phenotyping and ribotyping with strains isolated during the study showed that only TNNIII7 and TNMB5 are similar to the pathogenic strain TNEMF6....

Vibrio crassostreae sp nov., isolated from the haemolymph of oysters (Crassostrea gigas) ArchiMer

Polyphasic analysis of five new Vibrio isolates originating from the haernolymph of diseased cultured oysters is described. The new isolates were closely related to Vibrio splendidus, having 98 % 16S rRNA gene sequence similarity. gyrB phylogenetic analysis, fluorescent amplified-fragment length polymorphism (FAFLP) fingerprinting and DNA-DNA hybridization experiments clearly showed that the new isolates form a tight genomic group that is different from the currently known Vibrio species. The Summer Syndrome is a new shrimp disease that has been affecting a shrimp growout farm in New Caledonia since end of 1997. Development of an antiseptic procedure to improve retention rates during the grafting process of black-lip oyster, Pinctada margaritifera ArchiMer

In French Polynesia, black pearl cultivation now takes place on about 30 of the 118 atolls or islands. This activity has largely replaced copra, coconut oil and fishing as a source of income. Around 1,000 pearl farms, generally small ones of less than 5 ha, are registered. The pearl farms are mainly owned by families. In 1998, about 7,000 persons were employed in theentire pearl industry, from production to retailing. In value, this activity represents the first exportation of French Polynesian products. From 1988 to date, exports increased exponentially before leveling. Starting with less than two kilograms in 1972, exports have now stabilized at Il tons of "black" pearls.

Real-time PCR assay for rapid detection and quantification of Vibrio aestuarianus in oyster and seawater: A useful tool for epidemiologic studies ArchiMer

Because Vibrio aestuarianus is known to cause serious infections in Pacific oyster Crassostrea gigas, a real-time PCR assay was developed targeting the dnaJ gene of this bacterium. Adherence assays and Slime production of Vibrio alginolyticus and Vibrio parahaemolyticus BJM

In this study we investigated the phenotypic slime production of Vibrio alginolyticus and Vibrio parahaemolyticus strains, food-borne pathogens, using a Congo red agar plate assay. Furthermore, we studied their ability to adhere to abiotic surfaces and Vero cells line. Our results showed that only V. alginolyticus ATCC 17749 was a slime-producer developing almost black colonies on Congo red agar plate. Adherence to glace tube showed that all V. alginolyticus strains were more adherent than V. parahaemolyticus. Only V. alginolyticus ATCC 17749 was found to be able to form biofilm on polystyrene microplate wells (OD570 = 0.532).
